The Level of TBA-Active Products, Reduced Glutathione and Activity of Glutathione-Dependent En-zymes in the Blood Serum of Rats after Long-Term Injection of Omeprazole and Simultaneous Injection of Omeprazole and Multiprobiotic «Symbiter® Acidophilic»

The aim of the study was to determine the level of TBA-active products, reduced glutathione and activity of glutathione-dependent enzymes in the blood serum of rats with long-term gastric hypoacidity evoked by omeprazole (OM) and after simultaneous injection of OM and multiprobiotic «Symbiter® acidofilic» concentrated (SyM). The methods. The study was conducted on 40 white non-linear rats (180-200 g) were randomly divided into 4 groups. Animals of the 1st group (control) during 28 days were injected with 0,2 ml of H2O intraperitoneally (i. p.) and 0,5 ml of H2O (per os). Animals of the 2nd group during 28 days were injected with SyM (limited company “O. D. Pro-lisok”) in dose 0,14 ml/kg (per os) and 0,2 ml of H2O i. p. Animals of the 3rd group during 28 days were injected with inhibitor of Н+-К+-ATPase OM (14 mg/kg, diluted in 0,2 ml of H2O, i. p.) («Sigma-Aldrich», USA) and 0,5 ml of H2O (per os). The rats of the 4th group during 28 days were injected with the same dose of OM and SyM (0,14 ml/kg, per os). In a day of last injection of drugs in the blood serum we investigated the level of TBA-active products, reduced glutathione (RG) and glutathione-dependent enzymes: glutathione peroxidase (GP), glutathione transferase (GT) and glutathione reductase (GR). The results. Inhibition of gastric acid secretion evoked by OM injection to the rats during 28 days evoked the increase of TBA-active products in the blood serum by 88% (p≤0,05) in comparison with control. It is witness of activation of lipid peroxidation that it can be connected with disturbance of pro-/antioxidant balance in condition of dysbiosis development in gastrointestinal tract. Simultaneous injection of OM and SyM prevented the increase of TBA-active products in the blood serum. Injection to the rats during 28 days of OM diminished the RG level by 16% (p≤0,05), enhanced the activity of GP by 40% (p≤0,05), increased the activity of GT by 32% (p≤0,05) and decreased the activity of GR by 14% (p≤0,05) versus control. Simultaneous injection of OM and SyM prevented the diminishing of the RG level, the increase of GP activity and the decrease of GR activity. The introduction of SyM to the rats in condition of long-term hypoacidity led to decrease of GT activity by 39% (p≤0,05) in comparison with control, that in condition of normalization of lipid peroxidation process and RG level may be witness of modulating influence of SyM which decrease the GT activity also at separate injection to the intact animals. Conclusions. Long-term inhibition of gastric acid secretion evokes the increase of the level of TBA-active prod-ucts and the changes in functioning of glutathione link of antioxidant system in the blood serum of the rats. Multi-probiotic «Symbiter® acidofilic» concentrated has modulating influence on the level of lipid peroxidation and glu-tathione-dependent link of antioxidant system in the blood serum of the rats after long-term gastric hypoacidity.
