THE LEVEL OF IL-1Β, IL-10 AND IKBA EXPRESSION IN CHILDREN WITH CHRONIC GINGIVITIS SUFFERING FROM CHRONIC GASTRODUODENITIS

Aim. To study the level of pro- and anti-inflammatory IL-1β and IL-10 in the oral fluid of children with chronic gastroduodenitis, in relation to IkBα expression level. Materials and methods. The oral fluid and gingival scrapings of 50 children aged 6-12 years were studied to determine the level of IL-1β, IL-10, and IkBα; the children were divided into 3 groups. Group 1 included 10 children with healthy periodontium and no general medical conditions. The 2nd group had 20 physically healthy children with chronic catarrhal gingivitis. The 3rd group comprised 20 children with CGD and chronic catarrhal gingivitis. The mRNA expression of the IkBα gene and the content of IL-10 and IL-1β in the oral fluid were determined using real-time PCR. Results. According to the research findings, the content of pro-inflammatory IL-1β in the oral fluid of primary school children had different levels depending on their dental and physical health. It was the lowest in physically healthy children with no signs of periodontal inflammation. It was found that the content of IL-10, which has anti-inflammatory properties, was inversely proportional to the IL-1β concentration in the oral fluid of the children of the examined groups. The analysis of IkBα expression revealed its suppression in children of the second and third groups compared to healthy children. Conclusions. The results demonstrate changes in the IkBα levels in the gums of children with chronic catarrhal gingivitis; the impaired expression of IkBα may contribute to the deregulation of NF-κB pathways in the pathogenesis of periodontium inflammation. Decreased IkBα expression may specifically affect cytokine production and the inflammatory response associated with chronic catarrhal gingivitis accompanied by chronic gastroduodenitis.
