INFLUENCE OF THE COMPOUNDS CONTAINING OF SELENIUM ON THE HEAT SHOCK PROTEINS’ LEVEL AND THE MARKERS OF THIOL-DISULFIDE SYSTEM IN THE RATS’ BRAIN TISSUE IN ACUTE CEREBROVASCULAR INSUFFICIENCY

The compounds containing selenium have organoprotective, antioxidant action in the damage of different systems and organs including the brain tissue. Previously it was stated the protective metabolitotropic action of sodium selenite, cysteinum selenite, methioninum selenite on the markers of energetic metabolism, prooxidant-antioxidant homeostasis in the rats organs in toxic hepatitis. Also it was shown sodium selenite role in the thiol-disulfide system protection in acute cerebrovascular insufficiency. The changes of the thiol-disulfides markers was connected with the nitrogeniumoxide transport’ damage, with peroxynitrite formation, with oxidative and nitrosive stresses development. Early it was stated the influence of the compounds containing selenium on the enzymic link of the thiol-disulfide system in the rats’ brain tissue in acute cerebrovascular insufficiency – the activity of glutathione reductase, glutathione-S-transferase, glutathione peroxidase. The compounds containing selenium restored the activities of antioxidant enzymes. Aim of the investigation – estimate influence of the compounds containing selenium on the heat shock proteins’ level and the markers of nonenzymic link of thiol-disulfide system in the rats’ brain tissue in acute cerebrovascular insufficiency. Object and methods. The experiments were conducted on 40 male Wistar rats. Bilateral occlusion of common carotid arteries was performed under general thiopental-sodium anesthesia. In the rats’ brain tissue accordingly Methodical recommendation of the State Expert center of the Ukrainian Ministry of Health the content of nonenzymic markers of thiol-disulfide system have been identificated. There were investigated such markers as the contain of rebuilt intermediates (cysteine, methionine), markers of oxidative stress (homocysteine, nitrothyrosine), rebuilt and oxidative glutathione, rebuilt and oxidative thiol groups, the heat shock proteins’ level (HSP70). Sodium selenite in 50 mkg/kg, cysteine selenite and methioninum selenite in 30 mkg/kg have been administered intraperitoneally during 4 days in the case of identification of the thiol-disulfide markers and during 18 days in the case of investigation of the heat shock proteins’ level. Results. It was shown that in acute cerebrovascular insufficiency in the brain tissue on the 4thday the markers of nonenzymic link of the thiol-disulfide system have been changed. The content of cysteine, methionine, rebuilt glutathione, rebuilt thiol groups sum have been decreased. The level of homocysteine, nitrothyrosine, oxidative glutathione, oxidative thiol groups sum has been evaluated. In the 18th day of investigation the content of heat shock proteins’ level has been fallen also. The drugs containing selenium restore the content of the thiol-disulfide system markers of non-enzymic link on the 4th day and the heat shock proteins’ level on the 18th day. Cysteine selenite had the most expressive effect on all markers of thiol-disulfide system and heat shock proteins’ level. It was conjugated with the structure of cysteine selenite. Cysteine selenite contains of thiol groups, triazol ring and selenium. It may also influence on the activity of aerobic glycolysis and other markers of energetic metabolism expressively. Cysteine selenite changes the activity of the compounds on the dicarbonic and tricarbonic regions. So the compounds containing selenium have neuroprotective properties in the acute cerebrovascular insufficiency.
