DYNAMICS OF ENDOGENIC INTOXICATION INDICATORS CHANGES IN LABORATORY RATS OF DIFFERENT AGE GROUPS IN CONDITIONS OF POLYTRAUMA

The detoxification system is one of the body systems that undergoes significant changes in polytrauma. Violation of this system functioning leads to the development of endogenous intoxication (EI) syndrome which accompanies the diseases and complications associated with increased tissue decomposition, increased catabolism, internal organs insufficient function. To date, the age-related development mechanisms of EI are still unclear; there are no perfect pathogenetic approaches to explaining and predicting the early and late effects of polytraumatic lesions. The aim of the study – to investigate the content of EI markers – average molecular weight and the erythrocytic intoxication index value in animals of 3, 6 and 24 months of age at early and late time after the polytraumatic simulation. 72 non-linear white rats aged 3, 6 and 24 months were used in the experiment. Animals were modeled for severe trauma. The control group consisted of intact animals, which were kept under standard vivarium conditions. Deactivation of animals was performed under light, etheric anesthesia at the 1st, 3rd and 24th hour and 14th day of the experiment. Blood serum was used for the study. The toxic syndrome severity degree was estimated by the content of average molecular weight (AMW) and the endogenous intoxication index. As our studies have shown, the content of AMW undergoes significant changes. The content of AMW254 and AMW280 was increased at early and late terms after the polytrauma simulation in all age groups of animals compared to intact rats, indicating a pronounced increase in both the chain and aromatic amino acids in the peptide components of AMW. In our study, the content AMW of AMW254 in rats’ blood in three age groups was observed in early stages after the polytraumatic simulation, with a maximum value at the 24th hour. A similar dynamics of changes was observed in the study of AMW280 content in the blood of rats in three age groups of rats. An increase in the AMW content in the body of animals after the polytrauma simulation indicates in catabolic processes intensification. The AMW254 content increase, which may include oligopeptides, nucleic acid fragments, higher fatty acids, triglycerides, may indicate a violation of the hepatocyte membranes structure, and AMW280, which may be purine bases, uric acid and aromatic amino acids – the inhibition of the liver detoxification function. Simultaneously with the AMW accumulation in the blood of injured animals, the total toxic effect on the erythrocyte membrane also was increased, which manifested itself in a significant increase in eretrocytic intoxication index (EII) in all days of the experiment. The highest damage degree to erythrocytic membranes in rats of all studied age groups was observed at the 24th hour from the time of the polytrauma simulation and in 99 % in 3-months-old rats exceeded the level of intact animals. In the group of 6 and 24 months-old rats, this figure was 54 % and 45 %, respectively, higher than in intact rats. Consequently, the polytrauma simulation in animals of all ages was accompanied by an increase in both the content of AMW and EII value during all terms of the experiment. Probably these data indicate the catabolic processes intensification and the structure and function of cell membranes violation, which is accompanied by an increase in endogenous intoxication of the body. The features of changes in endogenous intoxication markers were close in their direction and severity, indicating toxic effects summation. A significant increase in the content of AMW and EII values we observed in young, non-sexually immature rats of 3 months of age at the 24th hour after the simulation polytrauma. In pubescent and old-age animals of 6 and 24 months of age these changes were less pronounced. Attention is drawn to the fact that on the 14th day of the experiment in animals of all studied groups, the content of AMW and EII value tended to decrease, although the level of the intact group of animals did not reach these rates.
