Infertility in marriage is currently quite common in the world and is a very big not only medical and biological, but also social and demographic problems. In recent years, many countries around the world have paid considerable attention to the male component of this pathological process. At the same time, it has been established that one of the causes of reproductive dysfunction in men is endocrine disorders. A serious problem in clinical practice is the crisis of the adrenal glands after the introduction of glucocorticoids. Objective: to investigate the nature of changes in the testes of rats with prolonged administration of high doses of prednisolone and after its abolition. The experiments were performed on rats given prednisolone intramuscularly at a rate of 0.4 mg/kg. The material for the study was collected 1, 3, 7, 14 and 28 days after drug administration and 1, 3, 7, 14 and 28 days after its withdrawal. The control group consisted of intact animals. The data obtained are fully consistent with modern ideas about the nature of hormonal effects on the sexual sphere and may be a consequence of the development of side effects that are embedded in Cushing’s syndrome. This includes an increase in animal body weight as a result of alimentary obesity, as well as an increase in the mass and volume of the testes with increasing their functional activity and a simultaneous absolute and specific increase in the number of immature sperm. With the sudden withdrawal of prednisolone reverse processes occur, and the decrease in the level of indicators is more intense than their previous increase. On the 14th day after weaning, the indicators become even significantly lower than the level of intact animals with simultaneous devastation of the lumen of the spermatic tubes, which may be the cause of infertility. In the future there is a partial recovery, although even on the 28th day after the abolition of prednisolone, some of them continue to remain significantly lower than the control values. With the use of high doses of prednisolone increases the weight and volume of the testicles of white rats with a simultaneous increase in the absolute and specific number of immature forms of sperm. On the 14th day after the abolition of prednisolone, the indicators become even significantly lower than the level of intact animals, which may be the cause of infertility, after which there is a re-restoration of the indicators towards the level of control values.
