CRISPR/CAS – АДАПТИВНАЯ ИММУННАЯ СИСТЕМА У БАКТЕРИЙ И ПЕРСПЕКТИВЫ ЕЁ ПРИМЕНЕНИЯ В РЕДАКТИРОВАНИИ ГЕНОМА

В данном обзоре литературы рассмотрены структурно-функциональные особенности и механизм действия системы CRISPR/Cas у бактерий, которая признана системой адаптивного иммунитета бактерий против инвазии вирусов и других генетических элементов. CRISPR/Cas состоит из определенного фрагмента ДНК, где повторы чередуются со спейсерами, в качестве которых выступают ДНК различных вирусов и плазмид. С CRISPR-блоком связаны СAS-гены. Они кодируют серию Cas-белков (в том числе Сas9) – полифункциональных ферментов с несколькими активностями (нуклеазной, хеликазной, полимеразной и др.), которые способны разрушать чужеродные ДНК. Природный Cas9 может применяться и в искусственных системах, но требует участия РНК-гида, который находит чужеродную ДНК-мишень благодаря комплементарности и нацеливает на нее Cas9. Последний приводит к деградации ДНК-мишени. Показана возможность применения CRISPR/Cas9-технологии в генной инженерии для получения геномодифицированных растений риса, сои, пшеницы, кукурузы, томатов, цитрусовых. Описаны попытки применения CRISPR/Cas для редактирования геномов при генетических, онкологических, сердечно-сосудистых, инфекционных заболеваниях.
